Modification of antibody class and binding properties typically requires cloning of antibody genes, antibody library construction, phage or yeast display and recombinant antibody expression. Here, we describe an alternative “cloning-free” approach to generate antibodies with altered antigen-binding and heavy chain isotype by mimicking the germinal center reaction in antibody-secreting hybridoma cells. This was accomplished by lentiviral transduction and controllable expression of activation-induced cytidine deaminase (AID) to generate somatic hypermutation and class switch recombination in antibody genes coupled with high-throughput fluorescence-activated cell sorting (FACS) of hybridoma cells to detect altered antibody binding properties. Starting from a single established hybridoma clone, we isolated mutated antibodies that bind to a low-temperature structure of polyethylene glycol (PEG), a polymer widely used in nanotechnology, biotechnology and pharmaceuticals. FACS of AID-infected hybridoma cells also facilitated rapid identification of class switched variants of monoclonal IgM to monoclonal IgG. Mimicking the germinal center reaction in hybridoma cells may offer a general method to identify and isolate antibodies with altered binding properties and class-switched heavy chains without the need to carry out DNA library construction, antibody engineering and recombinant protein expression. 相似文献

Neuroblastoma (NB) is a common pediatric cancer and contributes to more than 15% of all pediatric cancer-related deaths. Unlike adult tumors, recurrent somatic mutations in NB, such as tumor protein 53 (p53) mutations, occur with relative paucity. In addition, p53 downstream function is intact in NB cells with wild-type p53, suggesting that reactivation of p53 may be a viable therapeutic strategy for NB treatment. Herein, we report that the ubiquitin-specific protease 7 (USP7) inhibitor, , potently induces apoptosis in NB cells with an intact USP7-HDM2-p53 axis but not in NB cells with mutant p53 or without human homolog of MDM2 (HDM2) expression. In this study, we found that P22077 stabilized p53 by inducing HDM2 protein degradation in NB cells. P22077 also significantly augmented the cytotoxic effects of doxorubicin (Dox) and etoposide (VP-16) in NB cells with an intact USP7-HDM2-p53 axis. Moreover, P22077 was found to be able to sensitize chemoresistant LA-N-6 NB cells to chemotherapy. In an in vivo orthotopic NB mouse model, P22077 significantly inhibited the xenograft growth of three NB cell lines. Database analysis of NB patients shows that high expression of USP7 significantly predicts poor outcomes. Together, our data strongly suggest that targeting USP7 is a novel concept in the treatment of NB. USP7-specific inhibitors like P22077 may serve not only as a stand-alone therapy but also as an effective adjunct to current chemotherapeutic regimens for treating NB with an intact USP7-HDM2-p53 axis. The small chaperone protein Hsp27 confers resistance to apoptosis, and therefore is an attractive anticancer drug target. We report here a novel mechanism underlying the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) sensitizing activity of the small molecule , an inactive analog of the phosphoinositide 3-kinase inhibitor inhibitor LY303511, in HeLa cells that are refractory to TRAIL-induced apoptosis. On the basis of the fact that LY294002 is derived from LY303511, itself derived from quercetin, and earlier findings indicating that quercetin and LY294002 affected Hsp27 expression, we investigated whether LY294002 sensitized cancer cells to TRAIL via a conserved inhibitory effect on Hsp27. We provide evidence that upon treatment with LY303511, Hsp27 is progressively sequestered in the nucleus, thus reducing its protective effect in the cytosol during the apoptotic process. LY303511-induced nuclear translocation of Hsp27 is linked to its sustained phosphorylation via activation of p38 kinase and MAPKAP kinase 2 and the inhibition of PP2A. Furthermore, Hsp27 phosphorylation leads to the subsequent dissociation of its large oligomers and a decrease in its chaperone activity, thereby further compromising the death inhibitory activity of Hsp27. Furthermore, genetic manipulation of Hsp27 expression significantly affected the TRAIL sensitizing activity of LY303511, which corroborated the Hsp27 targeting activity of LY303511. Taken together, these data indicate a novel mechanism of small molecule sensitization to TRAIL through targeting of Hsp27 functions, rather than its overall expression, leading to decreased cellular protection, which could have therapeutic implications for overcoming chemotherapy resistance in tumor cells. An audit of the screening history of all new cervical cancer cases has been a requirement since April 2007. While NHS cervical screening programmes (NHSCSP) guidance requires that women diagnosed with cervical cancer are offered the findings of the audit, as yet there has been no research to investigate the psychological impact that meeting to discuss the findings might have on patients. This is in spite of the fact that cytological under‐call may play a role in as many as 20% of cervical cancer cases. This review draws on the literature concerning breaking bad news, discussing cancer and disclosing medical errors, in order to gain insight into both the negative and positive consequences that may accompany a cervical screening review meeting. We conclude that while patients are likely to experience some distress at disclosure, there are also likely to be positive aspects, such as greater trust and improved perception of care. 相似文献

SUMMARY In shallow lakes a siltation system develops when wind driven water turbulence erodes the lake floor, silt is resuspended and transported by means of waterdrift into the littoral vegetation. Here, due to the resistance of the vegetation, the turbulence is reduced and the silt deposited. Through this mechanism, the prevailing winds have a major rôle in the erosion and siltation pattern, lake morphology, vegetation pattern and in the mixing of water and nutrients from the open water and the reedswamp and vice versa. The model explains the uneven distribution of vegetation at the lakes Neusiedlersee (Austria) and Chilwa (Malawi) satisfactorily, if one assumes a positive correlation between siltation and reedswamp growth and spread. 相似文献

SUMMARY The length mass relationship for a Labeo capensis population is described by the equation M = 0,0134 L2'999 where 2,999 represents a constant the value of which indicates isometric growth. There is very little difference between the length/mass relationship for males and that for females and this is similar to the results found by Mulder (1973) for L. capensis in the Vaal River. The length/mass relationships for gravid males and females are different from that of the population as a whole indicating that maturity does result in a change of the body form which is in accordance with the results of Mulder (1973). A modified formula to take into account, small length intervals of an age group, is used to calculate the relative condition factor for Labeo capensis from the Caledon River for a consecutive “winter” and “summer” period. The results show that the conhd1t10ni for Labeo capensis of the 0+, 1+ and 2+ age groups is better in the summer period than in the winter period, whereas in the older age groups 3+ to 5+ the condition in winter is better than in summer. This can be attributed to the attainment of sexual maturity at the age of 3+ years and the presence of large quantities of fat deposited in the body during the winter months, which are then utilised for the development of the gonads prior to spawning. These results are similar to those obtained for the same species by Mulder (1973) in the Vaal River and by Bloemhof (1974) in the Hardap Dam. 相似文献

Aedes aegypti L. is the major vector of the arboviruses responsible for dengue fever, one of the most devastating human diseases. From a preliminary screening of fungal phytotoxins, cyclopaldic acid ( 1 ), seiridin ( 2 ), sphaeropsidin A ( 4 ), and papyracillic acid ( 5 ) were evaluated for their biting deterrent and larvicidal activities against Ae. aegypti L. Because compounds 1, 2, 4 , and 5 exhibited mosquito biting deterrent activities and 1 and 4 demonstrated larvicidal activities, further structure? activity relationship studies were initiated on these toxins. In biting‐deterrence bioassays, 1, 2, 4 , and 5 , 3,8‐didansylhydrazone of cyclopaldic acid, 1F , 5‐azidopentanoate of cyclopaldic acid A, 1G , the reduced derivative of cyclopaldic acid, 1 H , isoseiridin ( 3 ), 2′‐O‐acetylseiridin ( 2A ), 2′‐oxoseiridin ( 2C ), 6‐O‐acetylsphaeropsidin A ( 4A ), 8,14‐methylensphaeropsidin A methyl ester ( 4B ), and sphaeropsidin B ( 4C ) showed activities higher than the solvent control. Sphaeropsidin B ( 4C ) was the most active compound followed by 2A , while the other compounds were less active. Biting‐deterrence activity of compound 4C was statistically similar to DEET. In the larvicidal screening bioassays, only compounds 1 and 4 demonstrated larvicidal activities. Based on LD50 values, compound 4 (LD50 36.8 ppm) was significantly more active than compound 1 (LD50 58.2 ppm). However, the activity of these compounds was significantly lower than permethrin. 相似文献

Abstract The present study was carried out in abandoned fields in central Cyprus. The main objective was to examine the impact of the regeneration environment on the establishment and survival of Pinus brutia seedlings. Sixty-four permanent plots of 16 m2 were randomly established in two distinct sites. Four regeneration environments were recorded: (a) bare soil under the crown of a P. brutia tree, (b) soil under the canopy of a P. brutia tree and low shrubs, (c) bare soil in open areas, and (d) soil under the canopy of low shrubs in open areas. All P. brutia seedlings were classified in categories according to their regeneration environment. In all plots, the density of the P. brutia seedlings was measured in three different seasons (spring, summer, autumn). Soil temperatures were recorded, samples of surface soil were taken and the percentage of soil organic matter was measured. The main conclusions drawn from this research were the following: (1) the mature P. brutia trees and low shrubs facilitate the establishment and especially the survival of P. brutia seedlings, as all seedlings in bare vegetation ground had died by the end of the growing season, and (2) the importance of facilitation increases as abiotic stress rises. 相似文献

Abstract The first unambiguous documentation of hybridism in the genus Doronicum (Senecioneae – Asteraceae) is reported. All our morphological, karyological and molecular data concur to indicate that Doronicum × minutilloi Peruzzi hybr. nov. (2n = 60) is a hybrid growing in Monti Aurunci (Central Italy), originated from the spontaneous crossing D. orientale Hoffm. (2n = 60) × D. columnae Ten. (2n = 60). This new hybrid shows a slightly higher morphological, karyotypic and ribotypic affinity with D. columnae, but shares a trnL-trnF IGS haplotype with D. orientale, and co-occurs with the latter species only; it has reduced fertility and a high potential for vegetative propagation through rhizome fragmentation. Our results led us to suspect in fieri homoploid hybrid speciation. 相似文献
